You ever try Looney Tunes B-Ball for SNES?  That was a pretty good 2-on-2 game that somewhat addresses this topic.

Interesting,

So here's what I found after a really brief gameplay:

1) Animation is top notch, of course

2) Limited to just some WB characters (pros/cons)

3) I found the default button mappings awkward

4) Has an NBA Jam vibe to it, which makes sense given its Acclaim/Sculptured software background

5) The "wacky factor" adds the proper level of cartoon humor to it... and I'm glad it's adjustable!

Of course, in hindsight, plenty of things can be changed (more characters, bigger clock, etc.) but it could potentially make for a decent base/core/template for such a game idea.

Add it to the Jam!
